Understanding ISO 14644 Cleanroom Validation Standards
A practitioner's breakdown of particulate classification, recovery testing, and continuous monitoring obligations under ISO 14644-1:2015.

ISO 14644-1:2015 is the foundational classification standard for airborne particulate cleanliness inside cleanrooms and associated controlled environments. While the standard is often summarised by its Class numbers (ISO 5, ISO 7, ISO 8), the practical engineering effort lives in the testing protocols defined in ISO 14644-2 and -3.
Classification at three occupancy states
Every cleanroom must be classified under three distinct occupancy conditions — as-built, at-rest, and operational — because particulate behaviour fundamentally changes when machinery, gowned personnel, and active processes are introduced. Skipping the operational state is the single most common compliance gap we encounter during audits.

Recovery testing under ISO 14644-3
Recovery testing measures how quickly a cleanroom returns to its specified class after a deliberate particulate challenge. A well-engineered Grade B suite should recover within 15–20 minutes; failure here almost always points to undersized HEPA terminals or compromised room pressurisation.
- Particle count at-rest verified at minimum nine sample locations for rooms ≥10 m²
- Air change rates of 40–60 ACH typical for ISO Class 7
- Pressure cascade of ≥15 Pa between adjoining classified zones
- Continuous monitoring required for Grade A/B per EU GMP Annex 1 (2022)
